PP3010 Cryo-SEM/Cryo-FIB/SEM Preparation System

The PP3010 is a highly automated, easy-to-use, column-mounted, gas-cooled cryo preparation system suitable for most makes and models of SEM, FE-SEM and FIB/SEM.

The PP3010 has all the facilities needed to rapidly freeze, process and transfer specimens. The cryo preparation chamber is turbomolecular pumped and includes tools for cold fracturing, controlled automatic sublimation and sputter coating. After processing, the specimen is transferred from the cryo preparation chamber onto a highly stable SEM cold stage for observation. Cold trapping in the cryo preparation chamber and SEM chamber ensures the whole process is frost-free.

The Q Plus Series

The Q Plus Series is the latest version of Quorum’s internationally acclaimed, market-leading range of sputter and carbon coaters. They are increasingly used in high-resolution microscopy, where ultra-fine coating is a must.

Sputter coating is the standard method for preparing non-conducting or poorly conducting specimens prior to observation in a scanning electron microscope (SEM). GloQube® Plus Glow Discharge System for TEM Grids and surface modification

The GloQube® Plus is a cost-effective, compact and easy to use glow discharge system, designed to fulfil the needs of laboratories with TEM. The primary application of the GloQube® Plus is to modify the surface of TEM grids in a way that it meets requirements for successful imaging of a variety of macromolecules. The K850 combines versatility and ease of operation. Built-in thermo-electric heating and adiabatic cooling allow precise temperature control. The vertical pressure chamber (32 mm diameter x 47 mm) has a side viewing port, which gives a clear view of the liquid meniscus during filling. A magnetic stirrer ensures optimal mixing and exchange of process fluids.

Cryo-SEM: frozen hydrated root hairs

One of the most delicate structures borne on higher plants is the root hair. This is simply a projection of the outer tangential wall of the epidermis of the root, and has a mechanical function as well as a physiological one.

Because root hairs are over 90% water, conventional preparation methods, such as air or critical point drying, always result in considerable collapse, distortion and shrinkage of these structures. Low-temperature SEM (cryo-SEM), on the other hand, provides good preservation of structure without any collapse.

Root hairs illustrated here still have a thin layer of water (ice) on their surface, verifying their true frozen hydrated nature. If the ice proves to obscure important detail, it can be removed during the cryo-SEM process by careful sublimation (carefully raising the specimen temperature for a limited time).
